Her journey in acting began in Cuba with a leading role in the 2006 romantic drama Una rosa de Francia. At the age of 18, she relocated to Madrid, Spain, where she gained significant recognition for her role in the popular drama El Internado (2007–2010). Upon moving to Los Angeles, de Armas transitioned to English-speaking roles, appearing in the psychological thriller Knock Knock (2015) and the comedy-crime film War Dogs (2016). Her career truly took off with her performances as Joi, the holographic AI in the science fiction film Blade Runner 2049 (2017), and as nurse Marta Cabrera in the mystery film Knives Out (2019).

Her career continued to ascend with roles such as Bond girl Paloma in the James Bond film No Time to Die (2021) and her critically acclaimed portrayal of Marilyn Monroe in the biographical drama Blonde (2022). For her work in Blonde, she made history as the first Cuban actress to be nominated for the Academy Award for Best Actress. She is set to lead the action thriller Ballerina (2025), the fifth installment in the John Wick franchise. Her maternal grandparents were Spanish immigrants from the regions of Guardo, León, and Valverde de la Sierra, Palencia, both located in northern Spain. Her father, Ramón de Armas, had a diverse career, working as a bank manager, teacher, school principal, and even deputy mayor of a town. Notably, he had previously studied philosophy at a Soviet University. Her mother, Ana Caso, worked in the human resources department of the Ministry of Education.

De Armas has one older brother, Francisco Javier de Armas Caso, a US-based photographer. In 2020, he faced questioning by Cuban police due to his critical stance on Decree 349 and his connections to artists under government surveillance. Despite growing up with challenges such as food rationing, fuel shortages, and electricity blackouts during Cuba's "Special Period," de Armas has described her early life as a happy one.

During her childhood and adolescence, de Armas had no Internet access and limited exposure to popular culture outside of Cuba. She was permitted to watch "20 minutes of cartoons on Saturday and the Sunday movie matinee." Her family did not own a video or DVD player so she would watch Hollywood movies at her neighbor's apartment. She frequently memorized and practiced monologues in front of a mirror, and by the age of 12, she had decided to pursue acting. While a student, she filmed three movies. She left the four-year drama course just before presenting her final thesis to avoid the Cuban requirement for graduates to complete three years of mandatory community service before being allowed to leave the country. At 18, utilizing her Spanish citizenship inherited through her maternal grandparents, she moved to Madrid to launch her acting career.

Encouraged by her newly hired Hollywood agent, de Armas made the move to Los Angeles. When she arrived in 2014, she essentially had to "start her career again from scratch." She spoke very little English and often "didn't even know what [she] was saying" during early auditions. To overcome this, she dedicated four months to full-time English education, driven by a desire to avoid being limited to roles specifically written for Latina actresses. She made her Hollywood debut opposite Keanu Reeves in Eli Roth's erotic thriller Knock Knock (2015), learning her lines phonetically. While Randy Cordova of the Arizona Republic gave the film a positive review, he found de Armas's performance "unconvincing." Afterward, Reeves invited her to star in a Spanish-language role in Daughter of God, a thriller he also acted in and produced. Although producer Mark Downie hoped the film would be a star vehicle for de Armas, executive meddling severely edited the film, reducing her once-starring role. Frank Scheck of The Hollywood Reporter noted that while she was "appealing," de Armas couldn't fully demonstrate her "character's intense emotional demands."

De Armas next appeared in a supporting role in Todd Phillips's War Dogs (2016), portraying an arms dealer's wife opposite Miles Teller. Reviewing the film, Christy Lemire noted that while de Armas was "a hugely charismatic presence, she had little to do beyond functioning as the dutiful wife, save for a couple of showy moments." Her performance as Joi, the holographic AI girlfriend of Ryan Gosling's character in Denis Villeneuve's futuristic thriller Blade Runner 2049 (2017), earned significant praise. Mark Kermode of The Guardian lauded her for bringing "three-dimensional warmth to a character who is essentially a digital projection." Anthony Lane of The New Yorker found her "wondrous," noting, "Whenever Joi appears, the movie's imaginative heart begins to race." In May, de Armas had already confirmed she was engaged in multiple projects with Cruise, notably the upcoming Doug Liman-directed supernatural ocean thriller Deeper. An insider shared with PEOPLE earlier this month that the two actors possess a "special work relationship," noting de Armas' extensive "prep" this summer for Deeper, their inaugural joint film.

The source added, "Tom is exceptionally hardworking, and she's very enthusiastic about working with him. She speaks nothing but highly of him." De Armas's most recent romantic connection was reportedly with Manuel Anido Cuesta, the stepson of Cuban president Miguel Díaz-Canel Bermúdez. Before that, the Knives Out actress was linked to Tinder executive Paul Boukadakis.
